Mikado in the cell: Arrangement of proteins could be responsible for diseases NatureChemistry
-cut quartz plate as the local oscillator . These beams were collinearly passed through a 2-mm SrTiOplate for phase modulation and focused on the sample surface at an angle of incidence of 45°. The SFG signal from the sample interfered with the SFG signal from the LO, generating the SFG interferogram. The SFG interferogram was dispersed in a spectrometer and detected by a liquid nitrogen-cooled CCD camera .
We used SP measurements to study the protein interfacial adsorption, using a DeltaPi tensiometer and FilmWareX 3.62 software. The SP data were recorded versus time. SFG and SP experiments were performed simultaneously. The trough was cleaned with ethanol and then with water . Except when otherwise noted, the trough was filled with 4.8 ml of PBS buffer . The SP was also calibrated using pure water. The concentrated FUS LC protein solution was injected into the subphase with a glass syringe to reach a final protein bulk concentration of 1.5 μM. The sample was left to equilibrate until the SP stabilized.
