These enemies drop one blast powder more often than not. So, one night of skeleton killing should give you enough blast powder to use for the foreseeable future. However, should you need any more, you just need to repeat the above process a few more times. If you’re not near a desert biome, you can also head to the beach to take out pirate skeletons. These similarly only spawn at night and tend to be found in groups which makes killing them a little tricker. Furthermore, the drop rate tends to be a little worse than the cowboy pirates, so this method is a little less efficient.
Alternatively, you can find blast powder hidden around the map in chests. However, the spawn rate is completely random, and overall it’s the least efficient way of collecting blast powder. So unless you’re actively avoiding combat, killing the cowboy skeletons is the best way to go. The most efficient method is to to a desert biome, but failing that a beach biome works just as well.
