Increased TMPRSS2 expression in tongue tissue of females and alcohol drinkers identified as a potential risk factor for SARS-CoV-2 infection SARSCoV2 COVID19 coronavirus covid ACE2 tongue alcohol infection

By Dr. Chinta SidharthanOct 17 2022Reviewed by Danielle Ellis, B.Sc. In a recent study published in the Journal of Clinical Biochemistry and Nutrition, researchers explored the association between the expressions of angiotensin converting enzyme-2 receptor and transmembrane serine protease 2 in the tongue tissue and dysgeusia during severe acute respiratory syndrome coronavirus 2 infections.

About the study In the present study, the researchers used immunoblot analysis and immunohistochemistry techniques to determine ACE-2 and TMPRSS2 expression in the human and murine tongue tissue. Paraffin-fixed tissues were sectioned and mounted on glass slides. Immunostaining was performed using ACE-2 and TMPRSS2 antibodies. Tissues were homogenized and lysed using a lysis buffer after sonication, for the immunoblot analysis. Protein concentration was determined using a protein assay kit and sodium dodecyl sulfate-polyacrylamide gel electrophoresis . The immunoblots were incubated with ACE-2 and TMPRSS2 secondary antibodies conjugated with horseradish-peroxidase.

Additionally, the evaluation of ACE-2 and TMPRSS2 expression in other human organs revealed that ACE-2 expression is high in the kidneys, testes, and small intestine, whereas TMPRSS2 expression is upregulated in the breast tissue, liver, kidneys, and prostate gland.
